Plant tissue and organ imaging platform

Résumé

The plant tissue and organ imaging platform hosts important equipment and techniques for plant phenotyping, tissue patterning analysis or in situ localization of molecules such as nucleic acids and proteins in living organisms. On this platform, equipment for plant phenotyping can be used, with observations ranging from the macroscopic scale (photographic bench) to the microscopic scale for imaging of small objects using the GFP SZX12 binocular magnifier (microscope objectives X7 to X90-occular X10). Moreover, an intermediate and wide scale can be reached with the Keyence VHX-5000 3D digital microscope that allows to observe and reconstruct 3D images of biological objects larger than 50µm (microscope objectives X20 to X200 and X100 to X1000). On this platform, one can also run classical histology experiments to visualize plant tissue structures using different staining methods (toluidine blue, phlorogluvinol), as well as molecular histology techniques to study molecules of interest. Markers such as GUS can be used to localize gene expression patterns, or specific probes can be used for RNA, or antibodies for proteins. One can perform in situ hybridization or immuno-localization on whole-mounted or on sectioned tissues. For sections, we cut fresh tissue embedded in agarose using a vibratome, or fixed tissue embedded in paraffin or resin using a microtome. The observation of slides and the acquisition of images are performed with a Zeiss Axioscope microscope and analyzed with the Zen software. This platform is located in the PCV laboratory of the CEA, in the C2 building. It is coordinated by Christel CARLES, scientific manager, and Marie LE MASSON, technical manager.
