Improving transfer rate of antibodies on µarrays by using an acoustic transducer with low frequency vibrations

Résumé

During µarray building, the spotting mode leads to low efficiency in the ligand grafting and heterogeneities which limit its performances. To improve the transfer rate, we induced, with a home-made acoustic transducer, a hydrodynamic flow in every spotted droplet in order to disrupt the steady state which occurs during antibodies (Ab) grafting. Our acoustic device is a parallel micro-droplet mixer based on local acoustic field generation using low frequency vibrations. As a demonstrator, the transducer permits the simultaneous activation of 25 microdroplets localized on a micro-structured surface. Gold chips can be adjusted on the drop matrix leading to 25 well defined areas of contact. Intense mixing in each droplet was used to increase the transfer rate of antibodies in µarrays. By this way, Abs are expected to react strongly with the NHS activated gold chip surface. We have demonstrated with surface plasmon resonance imaging (SPRi) that such Ab-arrays fabricated with the activating mode present greater potential of antigens capture than standard chips. The mixing behaviour and the resulting rate of Ab grafting depend on a number of parameters namely volume, viscosity, surface tension, exciting membrane mode, and vibration velocity. We detailed extensive studies leading to optimized biochips performances allowing the reduction of ligands concentration and time consuming. Such effects are also expected during the biorecognition events with analytes and will be discussed.
