Study of electrochemotherapy effect in cell spheroids using a microfluidic device based on glass technology

Résumé

Electroporation (EPN) is a technique using pulsed electric fields to permeabilize and introduce molecules into cells without permanent damage. To assess the potential of cancer treatments based on EPN, like electrochemo- therapy (ECT), innovative in vitro assays exploiting 3D cell models such as spheroids are needed. The parallel treatment of tens of spheroids is required to produce statistical data. We designed a microfluidic device including a structured hydrogel placed between two conductive glass slides, enabling culture and electroporation of a large number of spheroids sharing similar characteristics. Here, we demonstrate its use to assess the effect of ECT on spheroids.
